Paper"}]},{"type":"Abstract","notation":[{"@value":"酵素活性の測定は, その測定条件つまり測定温度や溶液のpH, 濃度などに大きく左右される。そのため, 毎回同じ測定条件で測定できるように管理し確認すること, つまり精度管理は重要である。一般的に精度管理は酵素の標準物質を測定することで確認される。しかし, グルタチオンペルオキシダーゼ活性測定の精度管理は, 酵素の標準物質が不安定なため困難である。グルタチオンペルオキシダーゼは過酸化反応が関与している疾病を予防する機能を持っているので, その酵素活性を測定する意義は大きく, また精度管理も重要である。そこで, その測定に共役酵素として使用されるグルタチオンレダクターゼの希釈系列を調製し, グルタチオンレダクターゼの基質である酸化型グルタチオンとNADPHとで反応させ, そこでみられるNADPHの減少量をグルタチオンペルオキシダーゼ活性測定と同時に測定することで, 間接的ではあるが実用的なものと思われる精度管理の方法を検討した。その結果, グルタチオンレダクターゼの濃度とNADPHの減少量との間に, 比例関係を示す良好な直線性が認められた。さらに, その直線の傾きに高いくり返し精度(CV=2.9%)が認められたため,
