Experiments were carried out in 4 volunteers who have not been infected by hookworms for several years in the past, to study the problem of infection mode of hookworms. The larvae of both Necator americanus and Ancylostoma duodenale were allowed to act on the skin by placing them on the dorsal surface of feet and hands at the same time and in approximately the same number. The following observations were made: 1) The rates of penetration of larvae: 7.5-98.3% at 18℃ and 46.4-83.0% at 25℃ for A. duodenale; and 36.6-80.0 % at 18℃ and 62.1-62.5 % at 25℃ for N. americanus. 2) Dermatitis by Larvae was caused by N. americanus in 3 cases and by A. duodenale in 2 cases. Lasting for 1 to 5 weeks, it was attended by the subjective symptoms of itch and pain, and objective symptoms of red patches, papules, pigment deposits etc. 3) Hookworm infection was attended by subjective symptoms as follows: all the cases had cough, which was especially severe in 1 cases, persisting for a long period of time; 3 cases had expectoration; and 3 cases had hoarse voice, with 1 case almost losing her voice. 4) There was an increase in leukocyte counts and the number of eosinophil cells in 2 cases; the highest rates of increase were 40.0% and 38.5%, respectively. 5) Hookworm eggs were detected in the stools of 2 cases 66 days and 76 days, respectively, after the experiments were begun; and unfertilized eggs were detected in the stool of 1 case 56 days after the experiments were begun. The larvae of N. americanus alone were detected in the stools of 2 cases 66 days after the experiments were begun. 6) The rates of hookworm infection: 43.7 % for N. americanus and 4.1 % for A. duodenale in case 1: 10.4 % for N. americanus and 0 % for A. duodenale in case 2; 0 % for both N. americanus and A. duodenale in case 3; and 5.5% for N. amercanus and 0% for A. duodenale in case 4.
