To study the effect of Epo on proliferation and differentiation of UT-7, we maintained the UT-7 cell culture for more than 6 months in the presence of Epo. As a result, a subline, UT-7/Epo, was established. The growth of UT-7/Epo could be supported by Epo but not by GM-CSF or IL-3. UT- 7/Epo showed a greater level of heme content and ratio of benzidine- positive staining cells than did UT-7. Butyric acid promoted the synthesis of hemoglobin in UT-7/Epo, but not UT-7. Further, the mRNA concentrations of the c-myb oncogene and GM-CSF receptor beta-subunit were decreased substantially in UT-7/Epo cells. These findings showed that UT-7/Epo cells had progressed further in erythroid development than UT-7 cells, and suggested that long-term culture in Epo had promoted this differentiation. Whereas availability of the Epo receptor (Epo-R) for binding of Epo was reduced in UT-7/Epo cells compared with UT-7 cells, the Epo-R showed a similar affinity for Epo. This observation suggested that change(s) in postreceptor signaling step might be involved in the establishment and maintenance of the UT-7/Epo phenotype.</jats:p>"}]}],"creator":[{"@id":"https://cir.nii.ac.jp/crid/1381137044140214786","@type":"Researcher","foaf:name":[{"@value":"N Komatsu"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214659","@type":"Researcher","foaf:name":[{"@value":"M Yamamoto"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214657","@type":"Researcher","foaf:name":[{"@value":"H Fujita"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214660","@type":"Researcher","foaf:name":[{"@value":"A Miwa"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214656","@type":"Researcher","foaf:name":[{"@value":"K Hatake"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214661","@type":"Researcher","foaf:name":[{"@value":"T Endo"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214787","@type":"Researcher","foaf:name":[{"@value":"H Okano"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214784","@type":"Researcher","foaf:name":[{"@value":"T Katsube"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214785","@type":"Researcher","foaf:name":[{"@value":"Y Fukumaki"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]},{"@id":"https://cir.nii.ac.jp/crid/1381137044140214658","@type":"Researcher","foaf:name":[{"@value":"S Sassa"}],"jpcoar:affiliationName":[{"@value":"Department of Medicine, Jichi Medical School, Tochigi, Japan."}]}],"publication":{"publicationIdentifier":[{"@type":"PISSN","@value":"00064971"},{"@type":"EISSN","@value":"15280020"}],"prism:publicationName":[{"@value":"Blood"}],"dc:publisher":[{"@value":"American Society of Hematology"}],"prism:publicationDate":"1993-07-15","prism:volume":"82","prism:number":"2","prism:startingPage":"456","prism:endingPage":"464"},"reviewed":"false","url":[{"@id":"http://ashpublications.org/blood/article-pdf/82/2/456/228722/456.pdf"}],"createdAt":"2019-10-13","modifiedAt":"2019-11-20","foaf:topic":[{"@id":"https://cir.nii.ac.jp/all?q=Cell%20Biology","dc:title":"Cell Biology"},{"@id":"https://cir.nii.ac.jp/all?q=Hematology","dc:title":"Hematology"},{"@id":"https://cir.nii.ac.jp/all?q=Immunology","dc:title":"Immunology"},{"@id":"https://cir.nii.ac.jp/all?q=Biochemistry","dc:title":"Biochemistry"}],"relatedProduct":[{"@id":"https://cir.nii.ac.jp/crid/1390571395584246912","@type":"Article","relationType":["isReferencedBy"],"jpcoar:relatedTitle":[{"@language":"en","@value":"Looking Back on My Life of 40 Years as an Academic Person"}]}],"dataSourceIdentifier":[{"@type":"CROSSREF","@value":"10.1182/blood.v82.2.456.bloodjournal822456"},{"@type":"CROSSREF","@value":"10.14789/jmj.jmj21-0016-ot_references_DOI_HQsRIhtJef2Aa0PnPRRfgTsq4Ei"}]}
