Generation of Active Oxygen Species from Ischemic Heart Mitochondria and Effect of Vitamin Against the Oxygen Toxicity

Over the past 10 years, it has been growing to support the concept that ischemic stress produces the generation of active oxygen species and free radicals in the tissue. The ESR spin trapping technique was applied to heart mitochondria obtained from dog heart affected by ischemia, in order to detect the generation of active oxygen from the respiring mitochondria. ESR spectrum of active oxygen spin adduct was enhanced in ischemic mitochondria. The hydroxyl radicals were found to generate from mitochondria, by way of superoxide anion produced by electrons released from the electron transport system. The heart ischemia resulted in a decrease of membrane fluidity, with concomitant dysfunction of mitochondria and ATP depletion. The hydroxyl radicals were discussed to be the direct active oxygen species involved in the cellular damages. The antioxidant vitamins ; tocopherol, riboflavin, CoQ_<10>, were found to have the protective effect against the ischemic damages of heart mitochondria.
