HNA抗体の検出には抗原既知のパネル顆粒球が必要となるが, 顆粒球は長期保存が困難で, 血清によっては高い非特異反応を示し, HLA抗体とHNA抗体が混在する場合その判別が困難であり, パネル細胞としては最適なものではない. そこで我々は, HNA-1a, -1b, -2a抗体検出用パネル細胞として, 特定のHNAのみを発現し, 半永久的に使用可能な細胞株の開発を試みた.<br>【方法】HLA, HNAの発現がなく, 正常血清との反応性も極めて低いK562 (ヒト赤白血病細胞株) 細胞を標的とし, HNA-1a, -1b, -2a遺伝子をレトロウイルスベクターで同細胞に導入し, 遺伝子発現させ, その発現をそれぞれのHNAに対するモノクローナル抗体で調べた. さらに, 患者血清中に含まれるHNA抗体 (特異性既知) の同定もHNA-1a, -1b, -2aパネル細胞株を用いて行なった.<br>【結果】遺伝子導入した細胞は低バックグラウンドで高い特異性が確認された. また, 細胞を1年以上培養しても抗原発現の低下は見られなかった. さらに, HNA抗体以外の抗体 (HLA Class1抗体など) を含む患者血清でも特異的にHNA抗体を同定することが出来た.<br>【考察】作成したパネル細胞株は血中顆粒球抗体の高感度検出, とりわけ抗HLA
