説明

Siliac acid is a carbohydrate with 9 carbons, which is found on the terminal sugar chains of glycolipids and glycoproteins. It is known that sugar chains containing sialic acid play a role in intercellular signaling and in the recognition of cells and pathogens. N-acetylneuraminic acid and N-glycolylneuraminic acid (NeuGc) are the most widely detected sialic acids in mammals.
NeuGc is widely expressed in most mammals, but is not expressed in normal human or chicken tissues. NeuGc possesses immunogenicity in humans and chickens, and has attracted much attention as a tumor-associated antigen.
In this study, we performed the expression analysis of NeuGc in hepatocellualr carcinoma by immunohistochemistry method with chicken 2 monoclonal antibodies, Hu/Ch6-1 (against glycolipid type NeuGc), Hu/Ch2-7 (against glycolipid/glycoprotein type NeuGc).
In frozen liver sections from patients with hepatocellular carcinoma, glycolipid-type NeuGc was detected on the surface of liver cancer cells in 17 out of 37 samples (45.9%) by immunostaining using two chicken monoclonal antibodies against NeuGc and the tyramide signal amplification method. Increased serum levels of anti-NeuGc IgG and/or IgM were observed in 25 of the 37 patients with hepatocellular carcinoma (67.6%). The presence of these antibodies was mostly attributed to the expression of NeuGc on hepatocellular carcinoma cells. There was no correlation between serum AFP or PIVKAII levels and the presence of NeuGc or anti-NeuGc IgG and/or IgM.
We therefore consider that measurement of serum levels of anti-NeuGc antibodies is clinically meaningful and that anti-NeuGc antibody may be a useful screening test in combination with AFP and PIVKAII for diagnosis of hepatocellular carcinoma.
