Description

The effects of time from collection to seeding and deglycerol temperature on conception of frozen embryo in Holstein cows were examined. The morphologically normal embryos were frozen. After equiliblation in a glycerol solution (PBS + 0.4% BSA + 10% glycerol), each embryo was placed into a 0.25 ml plastic straw. After seeding at 5.1°C, the straws were kept at the same temperature for 15 min. And then, the straws were cooled at 0.5°C/min to -32.5°C before being plugged into liquid nitrogen. Glycerol (cryoprotectant) was removed by immersing embryos serially at 5 min intervals into 3 different 0.3M sucrose solutions containing 6, 3 and 0% of glycerol. The deglycerol treatment was conducted either at room temperature (1025°C) or on a warming plate (33°C). But pregnancy rate was not different between the 2 treatments. The pregnancy rate of frozen thawed bovine embryos kept less than 3 hr (65%) from flusing to freezing were significantly higher (P<0.05) than the rate of those kept over 3 hr (43%).
