Research Abstract

申請者が開発してきたFc-ARMを、抗体医薬を代替する医薬に高め、さらに抗体医薬の耐性の問題を克服することを目的とする。Fc-ARMのFc結合部として、大腸菌発現系で製造できる抗体ミメティックを採用し、高親和性のものを大規模ライブラリーから得る。こうして改良型Fc-ARMを作製し、その評価を行う。一方、耐性がんの問題を克服するFc-ARMの開発については、がんに共通の性質である腫瘍の酸性を認識して抗体を集積させるFc-ARMを開発する。そこで、腫瘍pHの認識部として、酸性pHに応答して細胞膜に挿入するペプチドを用いる。pH-ARMがNK細胞の性質を利用して、耐性の問題を克服できることを示す。
