Research Abstract
非閉塞性無精子症に対する根本的な治療法は存在しない。In vitroヒト精細管培養による精子形成の確立に向け、ヒト造精機能障害、特にearly maturation arrest症例について顕微鏡下精巣内精子採取術時に採取された精細胞の単一細胞網羅的遺伝子解析を行う。 １）分化・増殖が停止する細胞と正常な細胞の遺伝子発現の比較を次世代シーケンサーを用いて単一細胞レベルで行い、造精機能障害の分子生物学的メカニズムを明らかにする。 ２）Leydig細胞を含んだヒト精細管のin vitro培養を行い、培養条件の工夫により精細胞の分化・増殖を生じうるか、また培養後の精細胞の遺伝子発現を解析する。
