Research Abstract

ミトコンドリアtRNALeu(UUR)遺伝子上に位置する3243A>G変異は、重篤なミトコンドリア病を引き起こす一方で治療法は確立されていない。申請者は、3243A>G変異と3290T>Cの1塩基置換をもつmt tRNALeu(UUR) では、翻訳機能が回復しミトコンドリア機能が改善することを報告した。さらに、細胞外小胞(EVs)を介した3243A>Gと3290T>Cをもつmt tRNALeu(UUR)の補充がミトコンドリア機能回復につながる可能性を見出した。EVsの網羅的解析によりミトコンドリア機能改善に関与する成分を特定することで、ミトコンドリア病の新規治療法開発に貢献することを目指す。
