Research Abstract

高等真核生物は高度に発達したクロマチン構造をもつ染色体DNAを持っているが、DNA複製はこのクロマチンの上で行われる。これまでの研究から、DNA複製の進行中にはダイナミックなクロマチンの構造変換が起こることが分かっている。一方で、複製起点の確立に必要なクロマチンの構造変換の仕組みについては、ほとんど理解が進んでいない。本研究では、ヒトCdt1とその結合性因子が導くクロマチン構造変化を解析する。クロマチン上におけるDNA複製起点の形成反応を試験管内再構成し、クロマチン上でDNA複製起点が形成される過程を明らかにする。これにより、DNA複製起点の確立とクロマチンの状態変化についての知見を深める。
