Research Abstract

生体分子との結合において高分子のトポロジー形状は重要な役割を果たす。特に分子末端をもたない環状高分子は線形構造と比べて溶液中で取りうる分子コンフォメーションが制限される。これは結合に伴うエントロピー変化を軽減させ、標的分子との強い結合につながる。この原理は医薬の分野では環状ペプチドに適用されている。本申請では領域内研究として新たに「精密高分子」の技術を取り入れ、分子量および立体不斉が完全に決定された精密環状高分子を作製する。環状高分子の側鎖に糖鎖を修飾し、コレラ毒素タンパク質がもつ糖結合ポケットの放射状配置に対して合致する構造、およびそれに基づく強い結合を期待する。
