ふ卵14日目の鶏胚から調製した酸可溶性画分をセライトカラムクロマトグラフィーに供し,塩基類およびヌクレオシド類を分別後,Dowex1イオン交換カラムクロマトグラフィーにより,ヌクレオチド画分を得た。これを高圧濾紙電気泳動に供した後,L. leichmanniiを用いたマイクロバイオアッセイでデオキシリボチドを検出するとともに,分別した各デオキシリボチドを5'-ヌクレオチダーゼで分解し,生成したデオキシリボシドを検討した結果,酸可溶性画分中のデオキシリボチドは,dAMP,dCMP,dGMP,dTMPおよびdUMPと同定した。
Nucletide fraction in extract with cold perchloric acid from 14 day old chick embryos was prepared by successive column chromatography on celite and Dowex-1 ion exchanger. An aliquot of the nucleotide fraction was subjected to high voltage paper electrophoresis 3000 V for 90 min at pH 3.5, and then examined by microbioassay method with Lactobacillus leichmannii ATCC 7830. Four deoxyribotides (dCMP, dAMP, dGMP and dUMP or dTMP)were detected on the paper. Each compound on the electropherogram was eluted with water and digested with 5'-nucleotidase. The digest was developed on Tōyō No. 50 paper by ascending technique with n-butanol-water-conc. ammonia(255 : 45 : 0.5 v/v), and then investigated by the microbioassay. Deoxyriboside detected was dC, dA, dG, dU and dT, respectively. The results indicated that major deoxyribotides in the fraction were dCMP, dAMP, dGMP, dUMP and dTMP.
