The effect of voluntary running training on the myosin heavy chain (MyHC) isoform composition was investigated in the 17 and 19-week-old female Fischer344 rats (N=13). Animals were divided into two groups: sedentary (S; N=7) or voluntary running training (VT; N=6) group. Animals in group VT were given free access to a running wheel for 12 weeks. The single fibers were dissected from superficial portion of lateral gastrocnemius muscles (GAS). The single fiber type was determined by MyHC composition. The average running revolution of group VT was 6418±1117(Mean±SD) per day (Min.; 4837±1852, Max. ; 8340±2806). The body weight of group VT was significantly heavier than that of group S. The GAS weight of group VT was significantly higher than that of group S. There was no significant difference in the relative GAS weight in each group. In the relative composition of type IIB single fiber, the pure IIB fiber was decreased and hybrid fiber (included IIb+d MyHC) was increased. Hybrid fiber which included in type I MyHC was observed in type IIB hybrid fiber. In the relative composition of type IID single fiber, the pure IID fiber tended to decrease and hybrid fiber (included IId+b MyHC) tended to increase. However, there was no statistically difference between two groups. These results indicate that voluntary running training causes the increase of IId MyHC and increase in the relative composition of hybrid fiber, consequently.
