Abstract

Monoclonal antibodies against ochratoxin B (OTB) were generated by immunizing Balb/c mice with OTB conjugated to keyholelimpet hemocyanin (KLH). Eight stable hybridoma cell lines, named OTB.1, 2, 3, 4, 5, 6, 7, and 8, were obtained. All antibodies were IgG1・kappa. The detection limit was 0.01 ng/mL of OTB and 50 % binding inhibition was reached at 500 nM free OTB in an indirect competitive ELISA using OTA.2 antibody. It was approximately 1,000 times higher sensitivity than the previous reported. The cross‐reactivity of OTB.4 and OTB.5 antibodies were 25~46 % of ochratoxin A (OTA). OTB.2 and OTB.7 antibodies were equally reacted with OTA and B. OTB.1, OTB.3, and OTB.6 were strongly reacted with OTA (148~266 %) rather than OTB. The establishment of a high sensitive immunochemical assay such as ELISA or the immunoaffiny column method that were possible the detection of OTA and OTA by using these antibodies was expected.
