A hemorrhagic toxin from habu venom was separated into hemorrhagic factor-1(HR-1) and hemorrhagic factor-2(HR-2) based on the molecular weight of the protein. It is known that the natural inhibitor affecting the neutralization of HR-1 is present in the mongoose serum. However, this natural inhibitor has negligible neutralization activity against HR-2. Therefore, by using habu venom to carry out an immunity test in mongoose, we examined whether any component in the immune serum could be sufficiently effective in neutralizing both HR-1 and HR-2. The habu venom was administered into 10 mongooses for a period of 16 weeks. We drew blood 6 times of during this period and performed exsanguinations 16 weeks later. In this study, we used ELISA to measure the antibody titer for HR-1 and HR-2 in the collected serum. Further, we which we examined habu venom sensitivity in the 10 mongooses and the levels of anti-HR-2 antibody. We compared the results for each mongoose, and, found that there was considerable difference in sensitivity toward the habe venom in each mongoose. In order to evaluate the sensitivity, we used habu antitoxin, which was prepared at the Okinawa prefectural institute of health and environment, in horses and goats. To detect the immune serum component that effectively neutralizes both HR-1 and HR-2 we need to separate the immune serum into its functional components during further investigations, and examine the neutralization activity of the refined serum components in terms of the protein content.
