The presence of the antioxidant enzyme Cu/Zn-superoxide dismutase (SOD)and zinc in spermatogonia of Japanese eel has been shown to play an important role in the antioxidant system of germ cells. The purpose of this study was to clarify the mechanisms involved in the stress tolerance during oogenesis. The total SOD activity and the zinc levels, measured by a competitive inhibition assay with a tetrazolium salt, and zinc fluorescent probe, respectively, were found to decrease as the oocyte developed into more advanced stage. By employing an organ culture system, zinc deficiency induced by an intracellular zinc chelating agent (TPEN) was observed to cause atresia of follicle cells. Addition of zinc ions attenuates TPEN-induced atresia. These results imply a relationship between SOD and zinc, and suggests an involvement of zinc in stress tolerance of follicle cells.
