Male rats, weighing 300 to 350 grams, were nephrectomized bilaterally and their penis was ligated. The bladder was instilled with 0.7 ml of normal saline. This was designated as the intravesical fluid. Antibacterial agents such as kanamycin, DKB, gentamicin, cefamezin, rifampicin and nalidixic acid were administered to these rats 100 mg per 1 kg body weight. Forty-eight hours later, serum and intravesical fluid were submitted for thin-layer chromatographic bioautography. This procedure was made also at 24 hours after administration in the nalidix acid group. No active metabolite was detected in kanamycin, gentamicin, cefamezin and rifampicin. In DKB, serum sample occasionally showed the small inhibitory zone of a different Rf value from the original DKB, but an exact proof of being an active metabolite could not be obtained. In nalidixic acid, 24 hours samples, both serum and intravesical fluid, showed nalidixic acid as well as hydroxynalidixic acid. In 48 hours samples, both serum and the intravesical fluid, nalidixic acid could not be detected and only hydroxy-nalidixic acid was identified.
