The barley coleoptile cell-powdery mildew system is advantageous for observing host-parasite interaction. When a nonpathogen, Erkysiphe pisi, attacks coleoptile cells, cell level resistance (inaccessibility) is induced. The mechanism for recognition and the expression of inaccessibility were examined by this system. Cytoplasmic responses of coleoptile cells to attack by E. pisi were examined by the video-analysis. Coleoptile cells responded to E. pisi at the prepenetration stage and the fungus appeared to be recognized by coleoptile cells at that time. Confirmation of this point was attempted by cytological examination and the results from E. pisi germlings apparent by give out chemical signal(s) to bring about inaccessibility in coleoptile cells. Water-soluble extracts obtained from E. pisi conidia exhibited inaccessibility inducing activity. The cytological examination indicated the inaccessibility inhibitor which may suppress inaccessibility to possibly by released by the pathogen, E. graminis. Cytoplasmic responses of coleoptile cells at prepenetration stage indicated the actin cytoskeleton to parhaps be involved in the inaccessibility of coleoptile cells. When coleoptiles were treated with inhibitors of actin, E. pisi formed a haustorium. Actin cytoskeleton would thus appear likely involved in the recognition or expression of inaccessibility.
