Abstract

We constructed a cDNA library from a single developing rice grain. RNA was obtained from a sheet of manually peeled aleurone layer from one third of a single rice grain. Single stranded cNDAs were amplified using the PCR technique after oligo (dG) tails were introduced on the 3'-ends of the single stranded cDNAs obtained directly from the mRNAs. The resulting cDNAs were ligated into Bluescript SK+to obtain a cDNA library consisting of about 10^6 individual clones.
