Abstract

Cyclin‐dependent kinase‐like 5 (CDKL5) is a Ser/Thr protein kinase. Mammalian CDKL5 is predominantly expressed in brain and mutation in mammalian CDKL5 gene causes neurodevelopmental disorders. However, in fishes, functional role of CDKL5 is currently unknown. In this study, we investigated kinase activity and intracellular localization of zebrafish CDKL5. Two splice variants of zCDKL5, zCDKL5‐Long (zCDKL5‐L)(1081 aa) and zCDKL5‐Short (zCDKL5‐S)(1040 aa), were cloned from zebrafish adult brain cDNA library. In our previous studies, mammalian CDKL5 significantly phosphorylated amphiphysin 1 (Amph1) in vitro. Consequently, we cloned zAmph1 as a substrate of zCDKL5 from zebrafish embryo cDNA library. zCDKL5 colocalized with a part of zAmph1 in the cytoplasm of COS‐7 cells. Moreover, GST pull‐down analysis indicate that zCDKL5 bound to zAmph1. In addition, constitutively active zCDKL5(1‐350) efficiently phosphorylated zAmph1.
