Partial purification and some properties of the bacteriocin and the bacteriolytic substance produced by Clostridium perfringens strain K29

Description

Substances inhibiting the growth of sensitive cells and lysing them, produced by Clostridium perfringens strain K29 were separated in high purity by 50% ammonium sulfate precipitation, gel filtration on Sephadex G-100 and ion exchange chromatography on DEAE Sephadex A-50. The two substances obtained were compared for physicochemical properties and the effect on sensitive cells. At the range from pH 7 to 8, the inhibitory substance was stable, whereas the bacteriolytic substance was stable at the range from pH 5 to 8. Both substances were inactivated by proteinase and by heat treatment for 5 minutes at 50℃. The effect of the inhibitory substance appeared to be bactericidal but not bacteriolytic, whereas the bacteriolytic substance lysed sensitive cells rapidly. According to this investigation, it is satisfactory to consider the former as a bacteriocin and the latter as a lytic enzyme.
