The fluorescence properties of fluorescein conjugates of amylose and dextran were examined in aqueous solutions of pH7 as a function of the degree of substitution with dye, DS. The exitation and emission spectra of these conjugates were similar to those of free fluorescein and thus independent on DS. The quantum yield and excited lifetime were also close to those of free fuorescein except for the highest substituted conjugate. On the contrary, the degree of fluorescence polarization was dependent on DS even at low DS. the depolarization could be caused by both of the electric energy transfer and rotational diffusion of dye residues on the chain. The depolarization due to the energy transfer was interpreted in relation to the local concentration of the dye. It was found that the rotational relaxation time calculated using a modified Perrin equation was almost not dependent on DS, but the backbone structure of the polysaccharide chains.
