<jats:p>Natural infection of sand flies with <jats:italic>Leishmania</jats:italic> parasites was surveyed in an Amazonian area in Ecuador where leishmaniasis is endemic. Seventy-one female sand flies were dissected and one was positive for <jats:italic>Leishmania</jats:italic> protozoa. The species of this sand fly was identified as <jats:italic>Lutzomyia</jats:italic> (<jats:italic>Lu.</jats:italic>) <jats:italic>tortura</jats:italic> on the basis of morphologic characteristics. Analysis of the cytochrome <jats:italic>b</jats:italic> gene sequence identified the parasite as <jats:italic>L.</jats:italic> (<jats:italic>Viannia</jats:italic>) <jats:italic>naiffi</jats:italic>. We report the distribution of <jats:italic>L.</jats:italic> (<jats:italic>V.</jats:italic>) <jats:italic>naiffi</jats:italic> in Ecuador and detection of a naturally infected sand fly in the Ecuadorian Amazon and natural infection of <jats:italic>Lu. tortura</jats:italic> with <jats:italic>Leishmania</jats:italic> parasites in the New World.</jats:p>
