Identification of the nerve cell bodies with their axonal terminations in the central nervous system (CNS) is of first and foremost importance for neuroanatomists who are primarily concerned with studies of the neuropathway. Since the introduction of the methods of retrograde and anterograde tracing of axons, using horseradish peroxidase and isotope-labeled amino acids, respectively, these methods have become important tools for pursuing the neuronal connectivity. The new analytic techniques have produced some significant progress in this empirical science. However, misinterpretation of data due to careless observations may easily emerge, and this can be avoided by realizing the advantages and limitations of this approach as applied to the CNS. The present paper reviews the historical background of discovery of the "axonal flow", and describes some disadvantages of the axonal tracing techniques, which appear to have drawn insufficient attention of the investigators.
