Abstract
SSCP (single-strand conformation polymorphism) is widely used for mutation detection because of its simplicity and versatility. In this method, a segment to be searched for a mutation is amplified by PCR from genomic DNA or cDNA, denatured, and separated by electrophoresis in media having sieving properties without denaturant (1). Mutation usually causes a mobility shift in electrophoresis, and so, is detected. Conformational change of the single stranded mutant DNA is believed to be the reason for the mobility shift.
