Transcriptomic Analysis of the Innate Antiviral Immune Response in Porcine Intestinal Epithelial Cells: Influence of Immunobiotic Lactobacilli

Lactobacillus rhamnosus CRL1505 et Lactobacillus plantarum CRL1506 sont des souches immunobiotiques capables d'augmenter la protection contre les infections intestinales virales comme démontré dans des modèles animaux et humains. Pour mieux comprendre l'interaction hôte-immunobiotique, la réponse transcriptomique des cellules épithéliales intestinales porcines (PIE) à la provocation par le motif moléculaire associé au virus poly(I :C) et les changements dans le profil transcriptomique induits par les souches immunobiotiques CRL1505 et CRL1506 ont été étudiés dans ce travail. En utilisant la technologie des microréseaux et la PCR à transcription inverse, nous avons obtenu un aperçu global des gènes immunitaires impliqués dans la réponse immunitaire antivirale innée dans les cellules PIE. La stimulation des cellules PIE avec du poly(I :C) a significativement augmenté l'expression de l'IFN-α et de l'IFN-β, de plusieurs gènes stimulés par l'interféron, de cytokines, de chimiokines, de molécules d'adhésion et de gènes impliqués dans la biosynthèse des prostaglandines. Il a également été déterminé que les lactobacilles modulent différemment l'expression des gènes immunitaires dans les cellules PIE poly(I :C) -challengées. Les changements les plus notables ont été observés dans les facteurs antiviraux (IFN-α, IFN-β, NPLR3, OAS1, OASL, MX2 et RNASEL) et les cytokines/chimiokines (IL-1β, IL-6, CCL4, CCL5 et CXCL10) qui ont été significativement augmentés dans les cellules PIE traitées au lactobacille. Les immunobiotiques ont réduit l'expression des gènes IL-15 et RAE1 qui médient les lésions inflammatoires poly(I :C). De plus, les traitements par lactobacilles ont augmenté l'expression de PLA2G4A, PTGES et PTGS2 qui sont impliqués dans la biosynthèse de la prostaglandine E2. L. rhamnosus CRL1505 et L. plantarum CRL1506 ont montré des différences quantitatives et qualitatives dans leurs capacités à moduler la réponse immunitaire antivirale innée dans les cellules PIE, ce qui expliquerait la capacité plus élevée de la souche CRL1505 par rapport à CRL1506 à protéger contre les infections virales et les dommages inflammatoires in vivo. Ces résultats ont fourni des informations précieuses pour mieux comprendre l'interaction hôte-immunobiotique et son effet sur l'immunité antivirale. Les analyses transcriptomiques complètes ont identifié avec succès un groupe de gènes (IFN-β, RIG1, RNASEL, MX2, A20, IL27, CXCL5, CCL4, PTGES et PTGER4), qui peuvent être utilisés comme biomarqueurs prospectifs pour le criblage de nouveaux immunobiotiques antiviraux dans les cellules PIE et pour le développement de nouveaux aliments fonctionnels pour l'alimentation humaine et animale, qui peuvent aider à prévenir les infections virales.
Lactobacillus rhamnosus CRL1505 y Lactobacillus plantarum CRL1506 son cepas inmunobióticas capaces de aumentar la protección contra infecciones intestinales virales como se demostró en modelos animales y humanos. Para obtener información sobre la interacción inmunobiótica del huésped, se investigó en este trabajo la respuesta transcriptómica de las células epiteliales intestinales porcinas (PIE) al desafío con el patrón asociado molecular viral poli(I:C) y los cambios en el perfil transcriptómico inducido por las cepas inmunobióticas CRL1505 y CRL1506. Mediante el uso de la tecnología de micromatrices y la PCR de transcripción inversa, obtuvimos una visión general global de los genes inmunes involucrados en la respuesta inmune antiviral innata en las células PIE. La estimulación de las células PIE con poli(I:C) aumentó significativamente la expresión de IFN-α e IFN-β, varios genes estimulados por interferón, citocinas, quimiocinas, moléculas de adhesión y genes involucrados en la biosíntesis de prostaglandinas. También se determinó que los lactobacilos modulaban de manera diferente la expresión génica inmunitaria en células PIE expuestas a poli(I:C). Los cambios más notables se encontraron en los factores antivirales (IFN-α, IFN-β, NPLR3, OAS1, OASL, MX2 y RNASEL) y citocinas/quimiocinas (IL-1β, IL-6, CCL4, CCL5 y CXCL10) que aumentaron significativamente en las células PIE tratadas con lactobacilos. Los inmunobióticos redujeron la expresión de los genes IL-15 y RAE1 que median el daño inflamatorio poli(I:C). Además, los tratamientos con lactobacilos aumentaron la expresión de PLA2G4A, PTGES y PTGS2 que participan en la biosíntesis de prostaglandina E2. L. rhamnosus CRL1505 y L. plantarum CRL1506 mostraron diferencias cuantitativas y cualitativas en sus capacidades para modular la respuesta inmune antiviral innata en células PIE, lo que explicaría la mayor capacidad de la cepa CRL1505 en comparación con CRL1506 para proteger contra la infección viral y el daño inflamatorio in vivo. Estos resultados proporcionaron información valiosa para la comprensión más profunda de la interacción huésped-inmunobiótico y su efecto sobre la inmunidad antiviral. Lactobacillus rhamnosus CRL1505 and Lactobacillus plantarum CRL1506 are immunobiotic strains able to increase protection against viral intestinal infections as demonstrated in animal models and humans. To gain insight into the host-immunobiotic interaction, the transcriptomic response of porcine intestinal epithelial (PIE) cells to the challenge with viral molecular associated pattern poly(I:C) and the changes in the transcriptomic profile induced by the immunobiotics strains CRL1505 and CRL1506 were investigated in this work. By using microarray technology and reverse transcription PCR, we obtained a global overview of the immune genes involved in the innate antiviral immune response in PIE cells. Stimulation of PIE cells with poly(I:C) significantly increased the expression of IFN-α and IFN-β, several interferon-stimulated genes, cytokines, chemokines, adhesion molecules, and genes involved in prostaglandin biosynthesis. It was also determined that lactobacilli differently modulated immune gene expression in poly(I:C)-challenged PIE cells. Most notable changes were found in antiviral factors (IFN-α, IFN-β, NPLR3, OAS1, OASL, MX2, and RNASEL) and cytokines/chemokines (IL-1β, IL-6, CCL4, CCL5, and CXCL10) that were significantly increased in lactobacilli-treated PIE cells. Immunobiotics reduced the expression of IL-15 and RAE1 genes that mediate poly(I:C) inflammatory damage. In addition, lactobacilli treatments increased the expression PLA2G4A, PTGES, and PTGS2 that are involved in prostaglandin E2 biosynthesis. L. rhamnosus CRL1505 and L. plantarum CRL1506 showed quantitative and qualitative differences in their capacities to modulate the innate antiviral immune response in PIE cells, which would explain the higher capacity of the CRL1505 strain when compared to CRL1506 to protect against viral infection and inflammatory damage in vivo. These results provided valuable information for the deeper understanding of the host-immunobiotic interaction and their effect on antiviral immunity. The comprehensive transcriptomic analyses successfully identified a group of genes (IFN-β, RIG1, RNASEL, MX2, A20, IL27, CXCL5, CCL4, PTGES, and PTGER4), which can be used as prospective biomarkers for the screening of new antiviral immunobiotics in PIE cells and for the development of novel functional food and feeds, which may help to prevent viral infections.
