Publisher Summary Nervous tissues are extremely rich in glycoconjugates—particularly gangliosides—strongly suggesting important involvement of carbohydrate recognition in the nervous system. In this regard, it is of particular interest to note recent findings that certain gangliosides—when exogenously added to cultured cells—elicit various biological responses that seem to be closely associated with signal transduction pathway of the cells. Four types of such ganglioside-mediated responses are currently recognized: GM3-type responses, lectin/antibody/toxin-type responses, and glycoreceptor-type responses, glycosignals, cell nuclei targeting, and regulation of gene activity. Ganglioside GM3 modulates growth factor-dependent (EGF, PDGF, insulin) cell growth and associated receptor-mediated tyrosine phosphorylation. Whereas, lectin is known to affect growth and differentiation of cells, as seen from the blastogenic effect of concanavalin A on lymphocytes. There is an entirely different type of carbohydrate reaction that occurs within cell nuclei. Sialyl cholesterol (SC)—a synthetic amphipathic sialyl compound—has a potency to initiate and promote neurite outgrowth in mouse neuroblastoma cells, irrespective of its α or β form.
