抄録

The analysis of fat-soluble components such as tocopherols, free cholesterol, cholesteryl esters, and triacylglycerols in human plasma lipoprotein fractions was achieved by the reversed-phase high performance liquid chromatography. The plasma lipoprotein fractions- chylomicron, very low density lipoproteins, low density lipoproteins, high density lipoproteins 2, and high density lipoproteins 3- were collected individually by the ultracentrifugal flotation of human plasma. The chromatographic separation of hexane extracts of plasma or lipoprotein fractions was accomplished by the use of the column packed with Hitachi gel # 3057 which is a spherical octadecyl-silica having a particle size of 3-μpm. The mobile phase is acetonitrile-isopropanol mixture (75 : 25, v/v), and the eluate was monitored by an ultraviolet absorption detector and a fluorescence detector connected in series. Chromatographic peaks were analyzed spectrophotometrically and then identified by the field-desorption mass spectrometry after the peak collection. The high-resolution separation, detection and identification of the main fat-soluble components of lipoprotein fractions were performed by the methods mentioned above. In order to investigate the separation mechanism the authors analyzed the relationship between the structures of fat-soluble components and their capacity factors. Consequently, the authors confirmed that the separation of cholesteryl esters and triacylglycerols depends on the hydrocarbon chain Length and the number of double bonds of fatty acids which esterified those fat-soluble components. From the chromatograms, the authors demonstrated that each lipoprotein fraction has a characteristic profile and compositive data, and then the authors determined the contents of a-tocopherol, free cholesterol, and individual cholesteryl esters of individual lipoprotein fractions. Furthermore, the authors were able to make several conclusion about the localization of components in the steric structure of lipoprotein particles. As demonstrated in the results, our methods are actually useful for the diagnosis and treatment of many diseases such as cardiovascular diseases, atherosclerosis and hyperlipemia, and for the evaluation of the efficacy of drugs for these diseases, and for the studies in many other fields.
