Abstract

Determination of phenothiazine drugs in pharmaceutical preparations and in human seram by HPLC with electrochemical detection was carried out. To obtain highly sensitive detection, anodically oxidized glassy carbon electrode was set in the electrochemical detector. Chlorpromazine, promethazine, prochlorperazine and thioridazine were separated within 10 min on a cation exchange column Zipax-SCX using a mixture of methanol and 0.2 mol dm-3 borate buffer (pH 9.0), 0.5 mol dm-3 sodium nitrate(35 : 65) as an eluent. For each phenothiazine, a linear calibration curve was obtained ranging in amount from 100 pg to 10 ng and the detection limit was found to be ca. 30 pg. The recovery of phenothiazine drugs in pharmaceutical preparations by this method was over 98%. In the cases of serum samples, Extrelut(R) column was favorably used for extraction of phenothiazines from the test solutions. The results were satisfactory with a recovery of over 90% and a relative standard deviation of less than 1.9%.
