Abstract

A fluorescence method was developed for the estimation of immobilized ligands on a gel matrix. In the previous paper, we described an absorptiometric method. This method has various advantages compared with previously known methods, since it uses undestroyed gel beads as they are. The fluorescence method is also in this line, more sensitive and simpler in the procedure than the absorptiometric method. The proposed method used 0.06 % agarose solution to prepare a practically stable suspension of beads. The amount of ligands on beads was estimated by the fluorescence at the wavelength of maximum intensity. The amount of gel matrix in the suspension was estimated by the intensity of optical dispersion at 500nm. The present method was about twenty times as sensitive as an absorptiometric method. The method was applied to the measurement of the time course of immobilization reactions of naphthylethylenediamine ligand on polyvinyl gel matrix.
