Abstract

An improved method for rapid determination of enterotoxin-type of Staphylococcus aureus was developed. The procedure includes the following steps: a punched-out piece with a diameter of 8-10mm around a colony of S. aureus on the Baird-Parker medium was put in 0.5ml of phosphate buffer solution containing 0.5% bovine albumin and 2% rabbit serum. Enterotoxin (s) and coagulase extracted into the solution were detected by a reversed passive latex agglutination method using a “SET-RPLA” kit and by a micro-coagulase test, respectively.<BR>The new method showed good agreement with the usual shaking culture method: 95% in terms of enterotoxin producibility and 84% in terms of the type specificity of S. aureus. In the coagulase test, the degrees of agreement with the tube method using brain-heart infusion broth containing 5% rabbit plasma were 95% in terms of coagulase producibility and 100% in terms of coagulase typing.
