Abstract

An advantage of the spectrophotometer system based on a commercial high sensitivity spectrophotometer equipped with a flow through absorbance cell and an external microcomputer for the low-absorbance measurement is described. The use of a flow through absorbance cell (light pass : 100 mm) fixed in a cell compartment eliminates the errors caused by the exchange of absorbance cells. The smoothing of spectra and the correction of baseline drift performed on the external microcomputer greatly improve the quality of spectra. These contrivances made it possible to read out an absorbance of 0.0001 as a significant figure. Consequently, the sensitivity of this system was improved about 100 times compared to the usual methods. This system was applied to the spectrophotometric determination of iron (III) with Xylenol Orange, and a detection limit of 0.1 μg dm-3 was obtained.
