Color reaction of aromatic aldehydes and their related compounds with ο-phenyl phenol (OPP) was studied in sulfuric acid solution with the purpose to establish the detecting method of these compounds. It was found that aromatic aldehydes and their acetals gave a material with a color ranging from pink to reddish violet after the reaction with OPP in 50% sulfuric acid-methanol solution. This reaction was applicable to the detection of these compounds. Quantitative determination of these compounds was possible taking benzaldehyde and its acetals as the standards. Five milliliters of 50% (v/v) sulfuric acid methanol solution was added to the mixture of 100μl of the sample solution and 500μl of OPP solution (6×10-2mol/100ml methanol) in a 25ml test tube with a ground stopper. The test tube was kept in an ice bath during the mixing. After the mixture was heated for 70 minutes at 70°C in a water bath, the product was cooled in an ice bath. The absorbance at 523 nm of the solution was measured by using a 1 cm cell at a room temperature. By the proposed method, it was possible to determine (0.1?0.3)μmoles of the compounds in 100μl solution. Pigments and their precursors were isolated from the reaction product the column chromatography and the preparative thin layer chromatography, and analysed. According to the results of this analysis, it was found that the color of the reaction products was mainly attributed to the pigment of triphenylmethane-type compound, which was formed by the condensation reaction between 1 mole of aldehyde and 2 moles of OPP followed by the oxidation of the product.
