A new spectrophotometric method for the determination of 4-amino antipyrine (4-AA) was studied. 4-AA reacted with p-dimethylamino cinnamaldehyde (p-DACA) to develop intense color having an absorption maximum at 515 nm in ethanol with the addition of sulfuric acid. The relationship between absorbance at 515 nm and 4-AA concentration showed linear plots in the range of (0.62.5)μg/ml of 4-AA. The optimum conditions for the development were examined. To 1.0 ml of 4-AA sample solution {(0.6252.5) μg/ml}, 1.0 ml of 0.5% H2SO4-EtOH and 1.0 ml of 0.2% p-DACA-EtOH were added and made up to 10 ml of the total volume. The mixture was kept for 60 minutes in a water bath at 20°C. The absorbance was measured at 515 nm. The contaminant of aminopyrine did not interfere with the spectrophotometric method, but interfered with water contained in the sample solution. The coefficients of variation for the spectrophotometric determination were below 1% for (1.502.50) μg and below 6.2% for 0.50 μg of 4-AA.
