New diagnosis and treatment of lung cancer with tumor localizing Metalloporphyrins

Since 1978, we have examined a lot of porphyrin derivatives based on hemoglobin and chlorophyll and their metal complexes (500 in all), and reported the relationships between their side chain structures and tumor tissue accumulations. Porphyrin derivatives have been applied for photosensitizer, but it is necessary for using as diagnostic agents to regulate the side effects e.g. skin photosensitivity. Recently, we succeeded the regulation of photosensitivity by using of metalloporphyrin. We are now developing the metalloporphyrins for new tumor imaging radiopharmaceuticals (^<111>In-and ^<99m>Tc-ATN-10), new photosensitizers excited with YAG laser (ATX-70) and tumor-targetting anticancer agents (C12-Mn-Asp). ^<111>In-, ^<99>Tc-ATN-10 shows more clearer tumor images than those of ^<67>Ga citrate. In addition the photosensitivity of ATN-10 was not obserbed. ATX-70 shows severe tumor tissue destruction effects by irradiation of YAG laser based on hyperthermic, 2 photon absorption effect. Acute selective hemorrhagic necrosis of pancreatic adenocarcinoma implanted with Syrian golden hamsters was observed by administration of C12-Mn-Asp.
