Abstract

In order to establish an efficient ovule culture technique for producing hybrids between zonal geranium (Pelargonium×hortorum) and ivy-leaved geranium (P. peltatum), the effects of natural organic matter and growth regulators were investigated.<BR>Ovules were excised about 10 days after cross pollination and inoculated in modified White's agar medium (Abo-E1-Nil and Hildebrandt, 1973) for callus formation. For shoot regeneration, modified Linsmaier and Skoog medium (Reuther, 1982), supplemented with tryptone, various concentrations of 3-indoleacetic acid (IAA) and N6 (2-isopentenyl) adenine (2iP), and 3g/l gellan gum, was used. The highest regeneration rate was obtained when calli were primarily cultured in the modified Linsmaier and Skoog medium supplemented with 1500mg/l tryptone, 0.5mg/l IAA and 1.0mg/l2iP, and further subcultured in the same medium without tryptone. Shoot forming calli were cultured on a cooling plate in order to reduce hyper hydric shoots.
