Abstract

The formation of a down-growth of the junctional epithelium in periodontitis is believed to closely involve epithelial-mesenchymal interactions between the gingival epithelium and periodontal ligament, but many points remain unknown. In diseases presenting with tissue damage such as periodontitis, the involvement of matrix metalloproteinases (MMPs) has been suggested. Since matrix metalloproteinase-3 (MMP3) exhibits a strong level of substrate specificity to laminin-5 and type IV collagen (major components of the basal lamina), MMP3 may play an important role in periodontitis.<BR>Therefore, in order to clarify the mechanism of junctional epithelium down-growth, the present study investigated the actions of MMP3 by preparing a periodontal tissue model in which epithelial cell differentiation and proliferation could be reproduced using a three-dimensional culture system by periodontal ligament cells, gingival cells and gingival epithelial cells.<BR>The results showed that when cultured with a high level of calcium (0.97 mmol/l), epithelial cell layering and basal membrane-like structure formation were possible. Also, the layered epithelial cells expressed keratin. When cultured with a low level of calcium (0.075 mmol/l), epithelial cells maintained their proliferative potential and invaded the collagen gel, and MMP3 expression was observed in this area. The above findings suggest that the expression of MMP3 is strongly related to the mechanism of down-growth of junctional epithelium cells.<BR>Nihon Shishubyo Gakkai Kaishi (J Jpn Soc Periodontol) 51(2) : 153-161, 2009.
