The purpose of this study was to investigate the effects of bacterial endotoxin on bone resorption in vivo.<br>1. Endotoxin (lipopolysaccharide E. coli) mixed with penicillin and streptomycin was inserted into the root canals of dogs 7 days after pulpectomy. The animals were sacrificed 3 and 7 days after the application of endotoxin and the periapical tissues were examined histologically.<br>The experimental group showed marked cellular infiltration and bone resorption with numerous osteoclasts, but the control group showed little cellular infiltration or bone resorption. There were significantly more osteoclasts in the experimental group than in the control group (p<0.001).<br>2. Human peripheral blood lymphocytes were cultured in 2 media, one with endotoxin (100μg/ml), and one without endotoxin at 37°C in 5% CO2 and 100% humidity for 6 days. The lyophilized culture supernatants were inserted into the root canals of 2 groups of dogs. In a 3rd group, only lyophilized culture medium with endotoxin was used.<br>The pathological changes of the periapical tissues in response to the 3 different culture supernatants were similar in all 3 groups.
