Species identification of polychelid postlarvae collected from the Pacific coast of Honshu, Japan using DNA analyses

Abstract

Two unidentified postlarval specimens of lobster (Family Polychelidae) were collected off Fukushima and Suruga Bay, Japan in 2014. Species identification was based on DNA barcoding which provided partial DNA sequences of the mitochondrial COI and nuclear ITS 1 regions. DNA barcodes from 3 known adult species (Pentacheles laevis, Homeryon armarium, and Polycheles typhlops) were developed to provide comparative reference material to aid in species identification of the postlarvae. The DNA sequences of the postlarvae were very similar and were virtually an identical match (0-0.2% for COI region and 0.1% for ITS 1 region) to Pentacheles laevis. We therefore conclude that the species identity of both postlarval specimens to be Pentacheles laevis. This is the first known record from Japan of larval stage polychelid lobsters.
