Protective effects of Aspergillus oryzae NK-fermented grain germ on dextran sulfate sodium-induced colitis in mice.

【Objective】To estimate the protective efficacy of Aspergillus oryzae NK-fermented grain germ (A. oryzae NK-Koji) on inflammatory bowel disease, we examined the effects of the A. oryzae NK-Koji on pathophysiology and colonic mucosal cytokines of dextran sulfate sodium (DSS)-induced colitis in mice.<br>【Materials & Methods】Female Balb/c mice were randomly divided into 4 group (n=6/group). Mice in A. oryzae NK-Koji group were fed purified diet AIN-93M containing 2% or 5% the A. oryzae NK-Koji for 14 days before the induction of colitis and for 6 days thereafter. Colitis was induced by 3% DSS water administration ad libitum for 6 days. Mice in DSS-colitis control groups received the DSS water ad libitum for 6 days after the AIN-93M diet for 14 days. Normal control successively received the AIN-93M. On the end of the experimental period, the pathological findings of colonic tissue and the alteration of colonic mucosal cytokine levels were examined.<br>【Results】Administration of A. oryzae NK-Koji to DSS-ingested mice improved the body weight loss, the anus bleeding, the shortening of the colon length, the histological damage in the colon, and significantly reduced the overproduction of IL-6, IL-1β and TNF-α in the colonic mucosa of DSS-induced colitis in mice. Administration of the A. oryzae NK-Koji brought IFN-γ/IL-10 ratio in the colonic mucosa close to that of the normal control.<br>【Conclusion】A. oryzae NK-Koji demonstrated its protective efficacy on the pathological findings of the colonic tissue and the overproduction of inflammatory cytokines in the colonic mucosa of DSS-induced colitis in mice.
