Phosphoglucose isomerase (PGI), 6-Phosphogluconate dehydrogenase (6PGD), Phosphoglucose mutase (PGM) and Esterase-D (Es-D) isozymes were possible to detect simultaneously in the same agarose gel by electrophoresis in a continuous phosphate buffer system at pH 7.8. Electrophoresis was carried out at 16v/cm for 120 minutes followed by at 20v/cm for 10 minutes. The cooling (10°C) of the gel at electrophoresis was the most important in this method. The mixture of substrates for each isozyme in the same buffer (0.1 M Tris-HCI pH 8.0) was available to detect four isozymes simultaneously.<BR>The advantages of this method are as follow 1) 3 or 4 isozymes which have a similar optimal pH in a reaction are detected in one agerose gel simultaneneously. 2) More than 40 samples can be analysed in 2 or 3 rows on one agarose gel. 3) Only small amount of reaction mixture is needed (10 ml per 21 × 23 cm of gel) . 4) The time required for detection is short.
