抄録

Mannanase was extracted from the Koji culture of Rhizopus niveus and purified by column chromatographies on DEAE-Sephadex, SE-Sephadex, and Sephadex G-100. The crude extract contained mannosidase besides mannanase. These enzymes were effectively separated from each other by the Sephadex G-100 column chromatography.<br> The pH optimum of the purified mannanase was 5.5 and the temperature optimum was at 40°C.<br> The mannan from green coffee beans was hydrolyzed by the purified mannase and separated by a Sephadex G-15 column. The products obtained in high yield were di-, tri-, and tetrasaccharides, which consisted of only mannose and were not hydrolyzed by the enzyme.<br> An oligosaccharide containing mannose and galactose with a molar ratio of 6:1 was obtained in a small amount from the hydrolytic products by mannanase. This oligosaccharide had a polymerization degree of 7 and gave only galactose upon the hydrolysis with a hemicellulase of Bacillus subtilis.
