For the purpose of determination of asparagine and glutamine in fibroin and sericin of the silkworm, amino acid analysis by enzymic hydrolysis with alkaline proteinase and aminopeptidases was carried out. The total yields of amino acid by enzymic hydrolysis were low and some amino acids showed different mole percent from those by acid hydrolysis. Amino acid analysis with lithium buffers, however, clearly demonstrated the existence of asparagine and its amount in fibroin and sericin was approximately one half of aspartic acid. Trace amounts of glutamine were observed only in sericin. Trace amounts of tryptophan were also observed in both proteins by enzymic hydrolysis. By the with hydrolysis 3N mercaptoethanesulfonic acid, mole percent of tryptophan was determined at 0.24% in fibroin and 0.27% in sericin, respectively.
