Description
Using [1-13C] [2-13C] [1, 2-13C] sodium acetates as a precursor, the biosynthesis mechanism of Bombyx mori silk fibroin was examined with 13C NMR method. Time dependent 13C NMR spectra were observed on the hemolymph and silk fibroin prepared after these 13C labeled sodium acetates were given on 6th day of the fifth instar larva. These precursors were incorporated into the tricarboxylic acid, TCA cycle, through acetyl CoA and then, the cyclic reaction occurs, which are able to monitor with such a 13C NMR method. Glutamine and glutamic acid used as amino-donors were produced within one cycle of the TCA cycle and citrate was stored as a main source for the synthesis of silk fibroin. Especially, the mechanism of the alanine residue synthesis of the silk fibroin through malate was clarified on the basis of the detection of the 13C-13C spin coupling pattern.
