説明

Effect of long-term cold storage of male pupae and moth on reproductive ability in the silkworm was examined by using artificial insemination. As the results of prolonged storage of male pupae and moths at 5°C, only 40% of pupae and 30% of moths were able to survive after 2 months of storage. However, the survived pupae could not emerge and the moths lost their mating ability. On the other hand, no significant difference was found in the number of eggs laid and fertilization rate between normal mating and artificial insemination with sperm collected from either cold-storage pupae or moths. This fact showed that the sperm collected from seminal vesicles of survived pupae and moths still has fertility even after 2 months cold-storage.<br>Furthermore, a new method for determination of comparative sperm fertility was developed using competitive fertilization between the sperm of two strains. That is, the same volumes of sperm from both ‘white egg 2’ and ‘Hitachi×Nishiki’ were mixed and the mixed semen inseminated artificially to females of ‘white egg 2’. The segregation ratio of serosal color of deposited eggs, normal (pigmented) to white, revealed competitive fertility between sperm of two strains. When the mixture of two types of non-refrigerated sperm was inseminated, the female moths deposited eggs in which the segregation of serosal color was in the ratio of normal to white, 9:1, showing that the sperm precedence was stronger than white egg. On the contrary, the segregation ratio of normal color to white was reduced to 4:6 when the sperm collected from cold-storage (2 moth, 5°C) pupae of ‘Hitachi×Nishiki’ were used for artificial insemination in combination with sperm of non-refrigated pupae of ‘white egg 2’. This result indicated that the sperm fertility is damaged by cold-storaged of pupae to some extent.
