We succeeded in establishment of the artificial insemination of the silkworm, Bombyx mori, using a special instrument. The instrument consists of capillary tube, N2-gas cylinder, pressure regulator, three-way electromagnetic valve and foot-switch. The each 4-6μl of sperm collected from vesicula seminalis of emerged male or from bursa copulatrix of female moth after copulation was injected to bursa copulatrix of virgin female moth. The inseminated female oviposited favorably fertilized eggs. Numbers of the laid eggs, the fertilized eggs and the fertility were not significantly different from those from females normally copulated with males. The hatchability of the fertilized eggs oviposited by inseminated females was almost the same as that from females copulated with males. An experiment with genetic markers demonstrated that the fertilization by insemination was not caused by parthenogenesis but by sexual reproduction. The addition of g. prostatica was reconfirmed to be essential for both activities of motility and of fertilization of spermatozoa. Although the fertility was still very low, the fertilized eggs were oviposited by females inseminated artificially with semen which had been stored in liquid nitrogen using cryoprotectant.
