Description
Egg-specific protein (ESP) which is identified as one of the major yolk proteins of the silkworm, Bombyx mori, is found to be synthesized only in developing ovaries. A cDNA expression library was constructed from ovarian poly (A)+ RNA and an Escherichia coli expression vector, pKEN 602, according to the vector-primer method, cDNA clones for ESP were selected by in situ colony radioimmunoassay using an anti-ESP serum. One of them, a 1.9kb cDNA insert, was identified as an essentially full-length ESP cDNA by the fact that the amino acid sequence predicted from the nucleotide sequence of 5′-terminus matched with the N-terminal amino acid sequence estimated chemically on purified ESP. Moreover, the nucleotide sequence predicts the presence of the signal peptide consisting of 18 amino acids at the N-terminus.
