Fluctuations of the eclosion hormone (EH) activity during hatching and the relationship between the EH activity and hatching behaviour of the silkmoth, Samia cynthia ricini, were investigated. The precocious adult eclosion of both pharate-adults of Bombyx mori and Samia cynthia ricini was elicited by the administration of EH extracted from Samia embryos. It was clearly demonstrated that the extracts of the Samia embryos exhibited the EH activity. Hatching occurred at 9 days after oviposition and the time of hatching lasted about 2 to 3hr after lights-on, when the embryos were maintained under a 16L-8D photoperiod regime at 25°C. EH titer of the newly hatched larvae was higher than that of the pharate-first instar larvae (1 day before hatching). To determine the localization of the EH activity in the embryos, EH was extracted from either a head segment or thoraco-abdominal segments during hatching were observed: the EH activity in the thoraco-abcominal segment abruptly increased by abour 2.4-fold at 15min after lights-on and decreased gradually, though the fluctuations of the EH titer in the head segment were limited. It is possible to consider that EH is involved in the induction of the hatching behaviour in the silkmoth, Samia cynthia ricini.
