The molecular motion of silk fibroin at various water content was studied by means of spin-prove techniques. ESR spectra of organic nitroxide radical, 4-hydroxy-2, 2, 6, 6-tetramethyl piperidinooxy, dissolved in silk fibroin were mesured as a function of water content from 0 to 200%. It was found that the molecular motion of spin-prove in silk fibroin did not vary uniformly with water content but three regimes were distingished. ESR spectra showed little change and were broad, and anisotropy triplet signal was of below 14% water content. The correlation time of spin-prove in silk fibroin was very sensitive to water content between 20% and 64%. Above 64% water content, the correlation time decreased gradually with additional water content. It seemed possible to describe some empirical stoichiomertric relation between water and silk fibroin structure. However, specific water contents, 14, 20, and 64%, were associated with changes in the behavior and thus distinguished regimes which might differ by the nature of the water-silk fibroin interaction.
